Nice effort everyone but no one has solved it yet.
I'd be lying if I said that it's been getting easier to come up with new clues.
By the way, I modified clue #42 to (hopefully) make it less confusing.
    EON/name/lines of dialogue.

    1) We have the blues to make you happy.
    2) Everything began normally but...
    3) ...unbeknownst to me, a plan was starting to take shape.
    4) It certainly pays to be courteous.
    5) Have you tried the chicken soup?
    6) They pulled the old switcheroo on me but I don't seem to mind.
    7) Imagine my surprise when it suddenly happened.
    8) It was a shocking case of mistaken identity.
    9) You can't have one without the other.
    10) Honey and I share the same passion.

    11) What's going on behind my back?
    12) I can think of a few people who would gladly pay Scaramanga's fee right about now.
    13) By my own admission, I'm a loser.
    14) Wow, this has caused quite an uproar.
    15) The solution seems rather obvious but an attempt was never made.
    16) This might remind me of Christmas if I was in a better mood.
    17) Look what you've done to me; I'm at a loss for words.
    18) You can't miss my name. And yet, somehow you did.
    19) The state of someone's mental health was questioned.
    20) I only made one request, which I didn't think was unreasonable.

    21) There are no second chances.
    22) A monarch looks down upon us.
    23) Despite my best efforts, I was unable to meet your demands.
    24) A violent threat definitely didn't help matters at all.
    25) Unfortunately, I didn't pass the test.
    26) I hope that I can still be useful to you in the future.
    27) You might as well call us the three blind mice.
    28) Oh, how I'm trying to remember my training now. What's the regulation to cover this?
    29) It's plain to see that you're not a southpaw.
    30) There's another name you might know me by.

    31) The third time's the charm for Bond.
    32) One person could really get behind that idea.
    33) It was a real landmark moment.
    34) You rang?
    35) I know exactly what to use to fill the empty spaces.
    36) Looks like your plans have been put on ice.
    37) It turns out that I was just a pawn in your little game.
    38) I didn't say everything that I wanted to say.
    39) Someone left with more than they originally had. Someone else left with less.
    40) Didn't anyone teach you not to meddle in the affairs of others?

    41) Please take your circus act elsewhere.
    42) After what I said, maybe I should travel about two hours northeast of here.
    43) Your stubborn refusal to reverse course was infuriating.
    44) It's women's lib. They're taking over!
    45) When the chips are down, just buy more chips!

Don't believe she has a name (which I suppose negates her, based on a few clues), but I'll try, anyway: the receptionist girl from Perla de las Dunas, the one that Medrano attempts to rape?

The person hosting the current game usually specifies it. At the top of @pachazo's list of clues, he has it listed that it's an EON official character, with a name, who has lines of dialogue. Sometimes people may play someone who doesn't have a name or may not speak, and that's usually noted.

    Major Boothroyd.

During our long round (the one that went on for months) some people suggested doing just that. But the person whose turn it was pointed out, and I believe rightfully so, that such tactics are missing the spirit of the game, not to mention negates all of the time and artistry that went into building the clues. I'd recommend against it.

Humour (noun UK, humor US): the ability to find things funny, the way in which people see that some things are funny, or the quality of being funny.

lighten up (I know I shouldn't presume to tell you when to lighten up)

I think that's Ade who I already guessed, although he's not on the Incorrect list.
