    I agree with Remington, and the conversation evolves from those differences, but I think it goes beyond that as the usual film device to show us friction between Bond and this woman, only to later (seemingly) resolve the tension when they get together. A brief Hollywood-type scene with witty dialogue and one-upmanship.

    Religion wasn’t why Vesper wasn’t Bond’s type. It was because she was single and thought highly of herself. Bond prefers bored married women who are easier to get because they are trapped with husbands who don’t interest them anymore.

    Religion IS part of her cover, it is explicitly said in the conversation. Stephanie Broadchester or whatever her alias is and Mr Beach or whatever Bond's alias is are very much in love, but Stephanie's parents (notice that Vesper herself has no living relative) are strict Catholics so for appearance's sake Stephanie and Mr Beach or whatever the name is will sleep in two separate bedrooms. This is to explain why two people in a relationship don't sleep together. This is when Bond makes that joke about religion getting in the way. He refers to Vesper's cover, not to Vesper herself. Then Vesper refers to herself when she says she will keep her door shut, so Bond shouldn't get himself any ideas. This is then that Bond makes the remark: well you're not my type anyway because you're single, referring to Vesper, not Stephanie.

    Well we don't know exactly the full content of the document. Even if the Catholic family is not on it, then it's Vesper adding to the cover. In any case, it has nothing to do with Vesper being particularly religious or Catholic herself : she says her (or rather, her alter ego's) family is strict Catholic.

    You're reading too much into it. Her parents didn't directly die the moment she was born. She didn't appease anybody but merely used that argument as a dismissal towards Bond when she thought he was or would be coming on to her. You can't get more obvious than that.
